Anther culture

Anther culture is a process of using cultured anthers to breed haploid seedlings

Anther culture technique was created by Guha and Maheshwari in 1964. It could be applied to breed haploids for more than 200 species including tomato, rice, tobacco, oats, geranium. Some advantages of this method are as follows:
-          Simple technique
-          In some species, cell division is easy even when pollen cells are not really mature
-          Pollen rate with good reaction to cultured medium is high
-          Haploids could be produced with large quantity and rapidly.
In experiments on using Datura innoxia medium, Pollen rate with good reaction to cultured medium reached 100% and more than 1000 callus could be produced from an anther in the optimum condition. The result could be obtained within 24 hours after cell division.
Some disadvantages of breeding haploids by anther culture are as follows:
-          For some species, many seedlings bred are not haploid
-          For cereals, few green plants but more albinos or mosaic plants are obtained;
-          Anthers to breed haploids are easily removed
In order to obtain desired results, it is necessary to have specific strategies.
